Enhancement of Mif Expression Following Treatment with TLR7/8 Agonist Resiquimod

摘要:Background:TLR7/8 agonist Resiquimod(R848) is an immune response inducer used as prophylactic agent for several infectious diseases,as well as anticancer agents and vaccinal adjuvant.Its influence on Th1,Th2 and other cytokines expression was demonstrated in several studies.R848 functions on MIF and MIF receptors expression are not well documented.The aim of our investigation is to evaluate the influence of R848 on the regulation of MIF.Methods and results:To investigate the influence of R848 on MIF expression,B-cell lines(n =2; B95-8 and IM-9)and Epithelial cell lines(n=2; D407 and CNE-2 Z) were stimulated with 6μg/ml R848.After 24 h stimulation,Cells were harvested,supernatants collected and MIF and its receptors expression were accessed by real time RT-PCR and ELISA.At gene level,MIF was significantly upregulated in both B-cells(B95-8 cells:P value = 0.003;IM-9 cells P value = 0.002) and Epithelial Cells(D407 cell:P value = 0.0001; CNE-2 Z cells P value = 0.0002);however MIF expression was more increased after treatment with R848(P value = 0.001) in cancer cell line CNE-2 Z compared to the normal cell line D407.Furthermore,R848 was found to increase MIF release at proteins level.MIF receptors were differentially regulated by R848; at least one of MIF-related receptors was upregulated following stimulation with R848 in each cell line:CXCR4 in B95-8(P value = 0.003),CD74 in IM-9(P value = 0.001),D407(P value = 0.01) and CNE-2 Z(P value = 0.0003).Conclusion:MIF and its receptors expression were found to be enhanced by R848 at genes and proteins level,suggesting a probable activation of MIF downstream signal pathway.Thereby the strategy of R848 in cancer therapy need to be carefully managed.
