Gonadal Soma-derived Factor(gsdf) Initiates Testicular Differentiation in Teleost Fish,Medaka

摘要:Medaka Dmy(doublesex/mab-3 on the Y chromosome) is the second male determining gene reported in vertebrates.Both mutations within dmy gene or the promoter region,either resulting in a disruption of DMY protein,or a dramatic decrease of dmy expression,ultimately caused an XY feminization.Transgenic intergration of BAC fragment which contains the region flanking dmy gene resulted in a complete XX masculinization,further demonstrating dmy as a master sex determining gene in medaka.Gonadal soma-derived factor(gsdf),as one of direct downstream genes under dmy regulation,expresses almost simultaneously with dmy in Sertoli cells during the period critical for sexual differentiation at the embryonic stage 36(St36).Here we verified the gsdf effects as the testicular initiator through approaches of transgenic-expression and gene targeting.Ectopic expression of gsdf-2 A-GFP driven by β-actin promoter,induced a testicular differentiation with normal sperm development from genetic XX individuals,indicating its capability of male initiation in the lack of dmy.On the other hand,a targeting disruption of gsdf using zinc finger nucleases(ZFNs) mediated genome editing approach.A 100% of XY females were developed in gsdf knockout(gsdf KO)medaka,further demonstrating the indispensible roles of gsdf in the testicular differentiation of medaka.Most gsdf KO XY females have abnormality of ovarian development with low fertility or even sterility,indicating the wild effects of gsdf on gonad development.In order to understand the mechanisms of testicular initiation,we explore the bio-function of gsdf via a comparison of relavent gene expression,serum sex steroids levels,and FSH-LH expression in pituitary-gonad axis of wildtype and mutant gsdf individuals in medaka.
