摘要:The recently discovered wheat grain storage cysteine-rich avenin-like proteins(ALP) are capable of forming intra- and inter-molecular disulfide bonds.Previous studies have already proven that supplementation of heterologously expressed type-a ALPs in base wheat flour improved baking qualities,transgenic studies of type-b ALPs further proved that ALPs have positive effect on dough properties.In the current study,five genes of type-a ALPs were identified and mapped to chromosomes 7AS,4AL and7 DS,with previous studies confirming the three type-b ALP genes mapped to the group 7 and 4 chromosomes as well,as opposed to a group 1 and 6location for the typical glutens.The coding gene sequence of TaALPa-7A1 was 522 bp long,TaALPa-7A2 was 516 bp long,TaALPa-4A was 546 bp long,TaALPa-7D1 was 507 bp long,TaALPa-7D2 was 516 bp long,both TaALPb-7A and TaALPb-7D was 855 bp long,and TaALPb-4A was 858 bp long.By now twelve alleles were identified for TaALPb-7A and four alleles for TaALPb-4A.Ten alleles were identified at the TaALPa-7A1 locus,six alleles were identified at the TaALPa-7A2 locus,and six alleles were identified at the TaALPa-4A locus.No allelic variations were found at TaALPb-7D,TaALPa-7D1 and TaALPa-7D2.Allele specific markers were designed to distinguish two alleles at TaALPa-4A and TaALPb-7A.Some allelic variations result in frame shift of the original genes,encoding no functional proteins,meanwhile mutations,duplications,del/ins also resulted in polymorphic variations.It is highly possible that this minor family of proteins forms a new class of seed-endosperm proteins important in defining the quality characteristics of wheat flour.Future work will be focused on gene expression of ALPs genes,structural and functional prediction,phylogeny analysis of the gene evolution,allelic composition and associated effects,and marker assisted screening of good quality wheat.
