High frequency of abnormal high/low molecular weight glutenin alleles and two novel Y-type high molecular weight glutenin genes in Chinese wheat landraces of the Yangtze-River region

摘要:High and low molecular weight glutenin subunits(HMW-GS and LMWGS) are important components of wheat storage proteins,which play an important role in the determination of end-use quality of common wheat.Chinese wheat landraces are an important resource for exploring novel HMW-GS and LMW-GS genes to improve the wheat baking quality.A total of 485 common landraces of bread wheat were collected from the YangtzeRiver region of China.Their high and low molecular weight glutenin subunit composition were analyzed by Matrix-assisted laser desorption/ionization time-of-flight Mass Spectrometry(MALDI-TOF-MS).Results about HMWGS showed that among all landraces tested,453 were homogeneous for HMW-GS,32 were heterogeneous,and 37 contained abnormal subunits.A total of 22 alleles were detected,including 3 at Glu-Al,13 at Glu-Bl and6 at Glu-Dl,respectively.Higher variations occurred at the Glu-Bl locus compared with Glu-M and Glu-D1.Glu-Alc(74.0%),Glu-B1b(40.4%),and Glu-DIa(84.9%) appeared to be the most frequent alleles at Glu-M,Glu-Bl and Glu-Dl,respectively.Two alleles( "null" and 1) at the Glu-Al locus,three allele compositions(7 + 8,70 E + 8,7 + 9) at the Glu-Bl locus,and two(2+ 12 and 5 + 10) at the Glu-Dl locus appeared to be the common types in the 485 landraces.Sixteen new alleles represented by abnormal subunits were identified at the Glu-Bl and the Glu-Dl locus.Analysis about LMWGS indicated that 17 alleles at three loci:Glu-A3,Glu-B3 and Glu-D3 were identified,resulting in 87 different allele combinations.Out of the 17 alleles detected at all the Glu-3 loci,five belonged to Glu-A3,seven to Glu-B3 and five to Glu-D3 locus.MALDI-TOF-MS indicated Glu-A3a/c was present in72.8%,Glu-A3 b in 8.4%,Glu-A3 d in 8.4%,Glu-A3 f in 5.2%and Glu-A3 e in 3.6%.Seven types of alleles were identified at the Glu-B3 locus:Glu-B3d/’\(25.5%),Glu-B3b(21.3%),Glu-B3c(16.9%),Glu-B3h(13.8%),Clu-B3f(8.4%),Glu-63a(8.2%),and Glu-B3g(5.2%).Five types of Glu-D3 alleles were detected:GluD3a(58.4%),Glu-D3c(22.6%),Glu-D3d(15.5%),Glu-D3b(3.3%) and Glu-D3f(0.2%).Four new alleles represented by abnormal MALDI-TOF spectrum patterns were identified at the Glu-A3 andGlu-83 loci.Two novel Glu-lDy HMW-GSs(designated as 1Dy12.6 and 1Dy12.7) were identified and cloned from two Chinese wheat landraces Huazhong830 and Luosimai.The lDy12.6 and lDyl2.7 subunits were deposited as the NCBInr Acc.No KR262518 and KR262519,respectively.The full ORFs of lDy12.6 and 1Dy12.7 were 2,022 bp and 1,977 bp,encoding for proteins of673 and 658 amino acid residues,respectively.Each contains four typical primary regions of HMW-GSs(a signal peptide,N- and C-terminal regions,and a central repetitive region).Their deduced molecular masses(70,165 Da and 68,400 Da) were strikingly consistent with those identified by MALDITOF-MS(69,985 Da and 68,407 Da).The 1Dy12.6 is the largest 1Dy glutenin subunits cloned in common wheat,containing longer repetitive central domains than other 1Dy encoded proteins up to date.In comparison
