摘要:Apicomplexan parasites of the genus Babesia,Theileria and Plasmodium are very closely related organisms.Interestingly,their mitochondrial(mt) genomes are highly divergent.Among Babesia,the species Babesia orientalis is only recently identified and specifically epidemic in the southern part of China causing severe disease to water buffalo.However,nothing is known about its mt genome.In order to characterize the mt genome of B.orientalis and elucidate its genetic relationships with other mt genomes of Apicomplexan parasites,B.orientalis mt genome was determined by conventional PCR and sequencing.The entire mt genome of B.orientalis is 5996 bp in length with a linear form.It contains three protein-coding genes including cytochrome c oxidase Ⅰ(coxl),cytochrome b(cob) and cytochrome c oxidase Ⅲ(cox3) and six rRNA large subunit gene fragments.Phylogenetic analysis based on amino acid sequences of coxl,cob and coxl+cob,respectively,revealed that B.orientalis fell into Babesia clade with the closest relationship to B.bovis.This result is in agreement to the phylogenetic analysis based on 18 S rRNA and heat shock protein(HSP70).Our finding suggests that the mt genome sequences are also useful for the evolutionary studies of apicomplexan parasites.
