摘要:The rhoptry-associated protein 1 gene(rap-1) was found in numerous Babesia spp.A1732 bp rap-1 homologue was successfully identified from B.orientalis genome sequence,by comparing with B.bovis rap-1.B.orientalis rap-1(Borap-1) contains a1434 bp single open reading frame(ORF),which encoding a 52 kDa putative protein.The deduced protein sequence shared 57%similarity with other members of the Babesia RAP-1 thus was designated as the B.orientalis RAP-1(BoRAP-1).Phylogenetic analyses showed that the BoRAP-1 was most close to B.bovis and B.ovis which maintained the similar topologies from trees based on 18 s rRNA and HSP70 sequences.The 78 kDa recombinant protein contained a 26 kDa GST-tag,was expressed in Escherichia coli BL21TM.Western blot showed that the B.orientalis positive serum specifically reacted with the rBoRAP-1,while the antiserum against rBoRAP-1 recognized the merozoite of B.orientalis in immunofluorescence antibody test(IFAT).These results suggested that BoRAP-1 protein might be a potential diagnostic antigen and vaccine candidate for B.orientalis.
