Chitinase-transformed Arabidopsis thaliana Facilitate the Infection of HearNPV in Digestive Track of Cotton Bollworm

摘要:Chitinase derived from Heliocoverpa armigera Nucleopolyhedrovirus(HearN PV) plays essential roles in insect liquefaction and horizontal transmission between hosts. Additionally,Chitinase can enhance infection efficiency and virulence of HearN PV. Chitin layer is an important structure in insect digestive track to protect it from injuries by intestinal contents. In this study, 6 lines of positive transformants were obtained via floral-dip. The relative expression level of Chitinase significantly varies in these 6 lines. Line A(highest expression level) and line D(lowest expression level) were employed to feed third instar larvae of Cotton bollworm with HearN PV(dose: 22 OBs per larva). Results showed HearN PV entered enterocyte significantly earlier in worms fed with line A. LT50 and LT90were significantly reduced when larvae treated with line A and Hear NPV. With worms feeding on line A, mortality reached 100% at 8th day.Meanwhile, worms on line D were all dead at 11 th day. Virus productivity in larvae on line D was significantly lower than them on line A. However, daily weight gain was not different in each treatment. Our study suggests that Chitinase-transformed plants can significantly enhance virulence of HearN PV.
