Development of Enzyme-Linked Immunosorbent Assays for the Determination of N1,N12-diacetylspermine in Human Urine

摘要：N1,N12-diacetylspermine（DiAcSpm） is a diacetylated derivative of spermine.Urinary DiAcSpm levels of various cancer patients had been found to be significantly higher than those in normal people,which suggested that DiAcSpm might be a potential broad-spectrum tumor marker.In this study,anti-DiAcSpm polyclonal antibody was obtained by immunization of rabbits.8 kinds of analogous or relative molecules were employed to estimate the specificity of the antibody.The direct competitive and indirect competitive ELISAs were developed.And the sensitivity of the two assays were 7.79 ± 0.48 ng/mL（n=3） and 4.73 ± 0.67 ng/mL（n=3）;the limits of detection of the two assays were 0.55 ± 0.07 ng/mL（n=3） and 0.22 ± 0.02 ng/mL（n=3）,respectively.The indirect competitive ELISA was used to measure DiAcSpm in healthy people urine,and the recoveries of spiked DiAcSpm ranged from 109.96 to 117.13%.
