Edaravone, a radical scavenger, enhances anti-tumor effect of CPT-11 in murine colon cancer

摘要:<正> Aim: CPT-11 induces apoptosis in cancer cells by inhibiting the activity of topoisomerase I. However, CPT-11 is also known to activate the transcription factor NF-κB in cancer cells. Activation of NF-κB in cancer cells has been shown to attenuate apoptosis induced by chemotherapy or radiotherapy. Therefore, in order to improve the efficacy of chemotherapy, gene therapy using a modified form of IκBαor a decoy of NF-κB has been performed in animals, and the effectiveness of such therapy has been documented. In the present study, we assessed the effects of edaravone, a free radical scavenger on: 1) CPT-11-induced NF-κB activation in colon cancer cells; 2) the level of apoptosis after CPT-11 administration; and 3) tumor growth and lung metastasis.
