摘要:Chronic pain is a major health problem cause the economic-lost all over the world.It often co-occurs with anxiety disorder in clinic and they exacerbate each other.Most previous studies just focus on the basic neurobiology of pain at the molecular and cellular levels,while mechanisms underlying interaction of chronic pain and anxiety remain largely unknown.Here,we show that novel neural circuits involving glutamatergic projections from the anterior cingulate cortex(GluACC) to GABAergic neuron in the ventral tegmental area(VTA).The GABAVTA neurons project directly to the dopamine(DA) in VTA(DAVTA).And the DAVTA neurons project directly back to GluACC.Inhibition of ACC-VTA projection by optogenetics can relieve allodynia and anxiety disorder in spared nerve injury(SNI) mice.Activation of DA neuron in VTA relieves allodynia and anxiety disorder in SNI mice.Inhibition of VTA-ACC projection by chemicgenetics can relieve allodynia and anxiety disorder in SNI model.In addition,activation of VTA-mPFC projection relieves anxiety disorder but not allodynia.These findings indicate that a novel projection circuit between ACC and VTA may mediate the comorbidity of anxiety disorder and chronic pain in a way.
