Optimization of Extraction Process of Polysaccharides from Fruiting body of Cordyceps militaris L.Link by Orthogonal Test

【摘要】 [Objective]The research aimed to optimize the extraction process of polysaccharides from fruiting body of cultured Cordyceps militaris L.Link by orthogonal test.[Method] Decocting extraction method,hydrothermal refluxing method and alkali extraction method were used to extract polysaccharides from fruiting body of cultured Cordyceps militaris L.Link respectively.[Result]The optimum conditions of decocting extraction method were as follows:solid-liquid ratio of 1:40,extraction time of 3.0 h and extraction times of 3,and the effects order of 3 factors on the yield of polysaccharides was solid-liquid ratio> decocting time> decocting times.The optimum conditions of hydrothermal refluxing method were as follows:the sample powder was mixed with water at ratio of 1/20 and refluxed for 1.0 h,2 duplicates,and the effects order of 4 factors on the yield of polysaccharides was extraction times> extraction time> extraction temperature> solid-liquid ratio.The optimum conditions of alkali extraction method were as follows:extracting solution of 0.7 mol/L NaOH,solid-liquid ratio of 1:8,extraction duration of 0.5 h and extraction times of 3,and the effects order of 4 factors on the yield of polysaccharides was extraction times> concentration of NaOH> solid-liquid ratio> extraction time.[Conclusion]The optimum extraction process of decocting extraction method,hydrothermal refluxing extracting method and alkali extracting method were obtained in this study,which provided reference for the further research and industrial production of fruiting body from cultured Cordyceps militaris L.Link.
