The "Dose-Time-Toxicity " Relationship of Mice Hepatotoxicity Caused by the Administration of Water-extract Component of FRUCTUS EVODIAE after Multiple Doses

【摘要】 [Objective]To research the " dose-time-toxicity" relationship of mice hepatotoxicity caused by the administration of water-extract component of FRUCTUS EVODIAE after multiple doses.[Methods]Mice were given water-extract component of FRUCTUS EVODIAE by gavage at different time points or in different doses. The situation of death and toxic reaction of mice were observed. The serum ALT and AST activities at the same time point after administration were examined. The liver,renal and viscera indexes were calculated. And the morphological changes of liver tissue were observed under optical microscope.[Results]ALT and AST levels in mice serum and liver tissue enhanced at 1 d after administration; hepatotoxicity became significant at 3 d after administration,and it remained at 7 d. Compared with normal group, 0.63 5.0g/kg dose range of the water-extract component of FRUCTUS EVODIAE could cause significant hepatotoxicity injury within 7 d of administration; ALT,AST,AKP and TBI enhanced; ALB reduced; liver weight/body weight enhanced; edema of liver cells in different degrees was detected,as well as the fatty degeneration and hyperaemia among tissues,which showed certain dose-toxicity and time-toxicity relationships. These changes became serious with the increase of dose. And different dose groups had significant differences with normal control group.[Conclusion] Administration of water-extract component of FRUCTUS EVODIAE after multiple doses would cause significant mice hepatotoxicity,showing certain " dose-time-toxicity" relationship of mice hepatotoxicity.
