Optimization of Explants and Hormone Combinations for Callus Induction of Apocynum venetum L.

【摘要】 [Objective]To investigate the effects of different media and hormone combinations on callus formation of apocynum.[Method] Using bud-containing stems,stems and roots of Apocynum venetum L as explants,6.5 g/L agar,27 g/L sucrose and four hormones (6-BA,KT,NAA,2,4-D) were added to three media (MS,B5,KC) separately,totally nine schemes designed finally.The induction and browning rate,growth potentials and inducement value of callus were adopted as investigation indices for optimizing the callus induction conditions.[Result]The parameters for callus induction of A.venetum were optimized as follows: bud-containing stems as materials,medium for callus induction as MS + 6-BA 0.6 mg/L + NAA 0.5 mg/L.Under the optimized conditions,the callus induction rate reached 100% .[Conclusion] This study provided reference for establishing the A.venetum L suspension cell lines and researching the secondary metabolites.
