Purification Technology of the Total Flavonoids from Polygonum knotweed L. by Macroporous Resins

【摘要】 [Objective]To optimize the purification technology of the total flavonoids from Polygonum knotweed L.by macroporous resins.[Methods]With rutin as the reference substance,the content of total flavonoids in P.knotweed was detected by aluminium nitrate coloration method.Taking static adsorption capacity and static adsorption rate as the investigation indices,the adsorption and desorption effects of four macroporous resins on the total flavonoids of P.knotweed were compared.The optimal macroporous resin was selected;and the adsorption and desorption conditions were researched.[Results]HPD-100 macroporous resin was most suitable for the purification of total flavonoids from P.knotweed.The purification technology was as follows:2.5 g/L sample concentration,2 BV/h sample flow rate;2.5 BV sample volume,70%ethanol as the eluant,2 BV/h elution flow rate,and 5 BV eluant dosage.Under this technology,the concentration of total flavonoids in the purified products was 55.4%;and the extraction rate of total flavonoids reached 83.2%(n=3).[Conclusions]HPD-100 macroporous resin showed good purification effects on the total flavonoids of P.knotweed.The optimized technology was reasonable,feasible and stable.
