Extraction and Purification Technologies of Total Saponins from RADIX ROSAE LAEVIGATAE

【摘要】 [Objective]To select the optimal extraction technology of total saponins from RADIX ROSAE LAEVIGATAE;and to purify the total saponins by macroporous adsorption resins.[Methods]With the content of oleanolic acid as the reference substance,the total saponins content in RADIX ROSAE LAEVIGATAE was detected by UV-Vis spectrophotometry.Single factor test was adopted.The effects of extraction time,solid-liquid ratio and ethanol concentration on the total saponins of RADIX ROSAE LAEVIGATAE were investigated.Orthogonal test was designed according to the test results of single factor test.Purification effects of different types of macroporous resins on the total saponins of RADIX ROSAE LAEVIGATAE were evaluated according to the properties of static and dynamic adsorption and desorption properties.[Results]The influencing degrees of factors affecting the extraction rate of total saponins were in the order of solid-liquid ratio>reflux time>ethanol concentration.The optimal extraction technology was 1 h reflux time,1∶10 solid-liquid ratio,70%ethanol concentration,and two extraction times with 1 h each time.NKA-9 macroporous resin had the optimal purification effects on the total saponins of RADIX ROSAE LAEVIGATAE,and the purity of total saponins enhanced from 36.8%to 62.9%,and the elution rate reached 87.8%.[Conclusions]This method was effective,simple and feasible.
